文献和实验11-脱氧皮质〔甾〕酮 11-deoxycorticosterone
C21 H30 O3 , 21-羟基孕烯 -4-二酮 -3, 20。也单称脱氧皮质〔甾〕酮,或缩写 DOC。是副肾皮质激素的一种,具有糖代谢作用。由副肾皮质细胞的微粒体的 21-羟化酶的作用,从黄体酮进行生物合成的。其乙酸酯缩写为 DOCA,应用于临床。由于在副肾皮质细胞的线粒体中的 11β -羟化酶的作用可生成皮质酮,所以通常也认为是一种中间体。
1、原理与方法 基本上和C4高压电泳及免疫固定技术一致,样本不需经神经氨酸酶和羧肽酶B处理,电泳时直接加10uL EDTA抗凝血浆。凝胶板规格为125mm×260mm×3mm,电泳时电压控制在85mA,时间3~4h或血红蛋白迁移至离加样孔5.5em处时,停止电泳。用1:4或1:2稀释的抗Df血清(AtlanticAntibodiesTATB,USA或上海长征医院产品)进行免疫固定。依照Mauff提出的BF定型图命名。 2、所需试剂 (1)巴比妥缓冲液 巴比妥34.56g,NaOH6.00g
