哺乳动物细胞中，双链断裂（DSB）的基因组DNA 是一种潜在的致命病变。现已确知DSB 的形成为导致H2A 组蛋白的磷酸化。具体来说，在DNS 双链断裂处形成DNA 灶的过程中，DNA 损伤诱导剂可以诱导组蛋白变体H2AX 的在Ser139 位点上的磷酸化。磷酸化的H2AX 有利于募集蛋白质从而帮助DSB 部位进行修复。在哺乳动物细胞中，磷脂酰肌醇3-激酶样蛋白激酶，如ATM，ATR、DNA-PKCs 蛋白，以及磷酸化的组蛋白变体，H2AX。
