概述

ErbB3 belongs to the structure relatived ErbB receptor tyrosine kinases family,which contains the epidermal growth factor receptor(EGFR)subfamily.ErbB3 is one of the alternate transcriptional splice variants producted by the ERBB3 gene.It has a neuregulin binding domain but not an active kinase domain.Therefore,it can bind to the ligand but not convey the signal.Until,the ErbB3 forms heterodimers with other EGF receptor which do have kinase activity,the signal pathway transmits again.ErbB3 is mainly producted by epithelial tissues and brain,and overexpressed in subset of human mammary tumors.It interacts with Neuregulin 1,PA2G4,PIK3R1 and RGS4.The rHuErbB3-f is the 20-190 a.a.fragment of ErbB3 and has the function of induce specific antibody production in vivo,hence to inhibit tumor cell growth.It has been classified into therapeutic cancer vaccine and used to treat early,medium and advanced or post-operative breast cancer patients with over-expression of ErbB2.
