背景介绍
NR4A2 belongs to the orphan nuclear receptor (NR) family referred to as NR4A family, which have been implicated in cell cycle regulation, apoptosis, inflammation, metabolism and more recently in carcinogenesis [PMID:22583411]. NR4A2 also has a role in the expression of several proteins that are necessary for the synthesis and regulation of DA, such as tyrosine hidroxilase, DA transporter, vesicular monoamine transporter 2, and cRET [PMID:22294735]. Act as a transcriptional regulator, NR4A2 is essential for the differentiation and maintenance of meso-diencephalic mdDA neurons during development. It is crucial for expression of a set of genes such as SLC6A3, SLC18A2, TH and DRD2 which are essential for development of mdDA neurons [PMID: 17184956].

浓度很高。可见该法操作简便、经济，不过，腹水中常混有小鼠的各种杂蛋白（包括Ig），因此在很多情况下要提纯后才能使用，而且还有污染动物病毒的危险，故而最好用SPF级小鼠。（1）材料：a. 成年BALB/c小鼠。b. 降植烷（Pristane）或液体石蜡。c. 处于对数生长期的杂交瘤细胞。（2）方法：a. 腹腔接种降植烷或液体石蜡，每只小鼠0.3-0.5ml。b. 7-10天后腹腔接种用PBS或无血清培养基稀释的杂交瘤细胞，每只小鼠5×105/0.2ml。c. 间隔5天后，每天观察小鼠腹水产生情况，如腹部
