• 外切酶活性会产生单链 3´ 突出端，促使互补的末端重复序列退火
• 连接酶活性能将组装后的 DNA 切刻处进行连接
最终形成双链闭合的 DNA 分子， 可以用于 PCR、RCA 或其它的分子生物学应用，以及直接转化到大肠杆菌。
