| 产品介绍 | IgM normally constitutes about 10% of serum immunoglobulins. IgM antibody is prominent in early immune responses to most antigens and is largely confined to plasma due to it's large size. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. Due to it's high valency IgM is more efficient than other isotypes is binding antigens with repeating epitopes (virus particles and red blood cells) and is more efficient than IgG in activiating the complement pathway. The gene for the mu constant region contains four domains separated by short intervening sequences. 大多数IgM类抗体是优球蛋白不溶于水,故可用双蒸水透析纯化IgM。对那些溶于水的IgM类抗体可用饱和硫酸铵沉淀。沉淀后可以采用颗粒排斥层析法(凝胶过滤)进一步纯化。 颗粒排斥层析(size-exclusion chromatography)法又称分子筛层析或凝胶过滤,是利用微孔凝胶分离不同大小分子的抗体,可用于样品中IgG和IgM的分离,常用于对硫酸铵粗提物的进一步纯化。通过该法纯化所得抗体可用于Ig分子片段的制备、FITC、生物素或同位素标记。 一、材料和试剂
(一)材料 1.血清 2.葡聚糖凝胶G200 3.洗脱液0.01Mol/L pH7.4PBS(0.14Mol/L NaCl或0.1Mol/L pH8.0 Tris-HCl 0.14Mol/L NaCl)。 (二) 操作方法 选取2.50cm×90cm层析柱,用葡聚糖凝胶G200装柱,平衡后,直接加血清样品10ml或硫酸铵粗提制品,洗脱液洗脱,流速0.25ml/min,分管收集,测OD280 值,第一峰即为IgM
