goodRIPA裂解液(RIPA的本意是Radio Immunoprecipitation Assay)是一种传统的细胞组织快速裂解液,主要用于从动物细胞和组织中提取可溶性蛋白,其裂解得到的蛋白样品可以用于常规的Western Blot、IP及Elisa等实验。RIPA裂解液的配方有很多种,根据其裂解强度大致可以分为强、中、弱三类。用RIPA裂解液裂解得到的蛋白样品,可以使用 BCA蛋白定量试剂盒 (货号:ZJ101或ZJ102)测定蛋白浓度。

go注意:如果样本裂解不充分,可以适当提高裂解液的用量;若需要高浓度的蛋白样品,也可适当降低裂解液的用量。

样本的获得。 Western Blot实验主要目的是检测蛋白质的表达,所以首先得从实验样本中获取蛋白质,目前使用较多的样本是细胞及哺乳动物组织、其次是细菌及真菌、再后是植物。 细胞及哺乳动物的组织:目前提取与纯化蛋白质最经典的方法是RIPA强、中、弱系列的裂解液及NP-40裂解液、SDS裂解液、哺乳动物蛋白抽提试剂,组织蛋白抽提试剂。这几种裂解液裂解原理相似,但作用效果不同,实验者需要根据自身实验需求来选取合适的裂解方式,例如RIPA(强)裂解液与SDS裂解液是作用效果很强的裂解液,可以使样本充分
?如果我想检测全细胞的蛋白,不分可溶性和不溶性,应该怎么做呢? waterzouyx RIPA裂解液也分强、中、弱的,要看看你的目的蛋白是否为难溶性蛋白,如果为难溶,你的沉淀中有目的蛋白也正常了。暂时没听说不溶性蛋白可以做WB的。个人认为你要求的全细胞蛋白的问题,你可以考虑用二维电泳用的蛋白裂解液,其裂解效率非常高,不溶性沉淀非常少 。另提醒下,RIPA法裂解的蛋白提取物,不能用Bradford法测定蛋白浓度的 zhangyong1980121
