脱氧核糖核酸酶Ⅰ(DNaseⅠ)是一种非特异性核酸酶切酶，大多数来源于重组E.coli菌株，含有牛胰腺DNaseⅠ的MBP融合克隆。DNaseⅠ可用于降解单链或双链DNA，其原理为DNaseⅠ水解磷酸二酯键产生带有5'-磷酸基团和3'-OH的单核苷酸或寡核苷酸。Mg2+或Mn2+都可以激活DNase I 的活性，而Ca2+浓度直接影响酶的活性。Mg2+存在时可在双链DNA的每条单链上随机产生切口；而在Mn2+存在下可使双链DNA断裂，使DNA片段化。
操作步骤(仅供参考)：
取DNaseⅠ(RNase free)平衡至室温，低速离心，使液体沉至管底待用。
根据不同实验，应加入适量的酶，以便充分消化DNA。
大多数情况下，取DNaseⅠ(RNase free) 2～5μl(即4～10U)，加入10×DNase Buffer 10ul以及待处理液，后用去离子水或RNase-Free ddH2O补至100μl。
25～37℃孵育10min。
灭活条件：75℃孵育10min。
