| 产品介绍 | background: CCR6 is a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein coupled receptors. It is preferentially expressed by immature dendritic cells and memory T cells. The ligand of this receptor is macrophage inflammatory protein 3 alpha (MIP 3 alpha). This receptor has been shown to be important for B lineage maturation and antigen driven B cell differentiation, and it may regulate the migration and recruitment of dentritic and T cells during inflammatory and immunological responses. CCR6 has been reported to be expressed in intestine, colon, liver, lymph node, spleen, lung, blood, umbilical cord, placenta, skin, and pancreas. It has also been reported to be expressed in lymphocytes, T cells, and B cells. Function: Receptor for a C-C type chemokine. Binds to MIP-3-alpha/LARC and subsequently transduces a signal by increasing the intracellular calcium ions level. Subcellular Location: Cell membrane; Multi-pass membrane protein. Tissue Specificity: Spleen, lymph nodes, appendix, and fetal liver. Expressed in lymphocytes, T-cells and B-cells but not in natural killer cells, monocytes or granulocytes. Similarity: Belongs to the G-protein coupled receptor 1 family. Primary Antibody:Rabbit Anti- CCR6 antibody(bs-1542R), Dilution: 1μg in 100 μL 1X PBS containing 0.5% BSA; Isotype Control Antibody: Rabbit IgG(orange) ,used under the same conditions ); Secondary Antibody: Goat anti-rabbit IgG-PE(white blue), Dilution: 1:200 in 1 X PBS containing 0.5% BSA. Protocol The cells were fixed with 2% paraformaldehyde (10 min) . Primary antibody (bs-1542R, 1μg /1x10^6 cells) were incubated for 30 min on the ice, followed by 1 X PBS containing 0.5% BSA + 10% goat serum (15 min) to block non-specific protein-protein interactions. Then the Goat Anti-rabbit IgG/PE antibody was added into the blocking buffer mentioned above to react with the primary antibody at 1/200 dilution for 30 min on ice. Acquisition of 20,000 events was performed. |
