应用

逆转录酶(RT)活性的测定已广泛应用于逆转录病毒繁殖的体外测试。早期原病毒DNA的合成需要RT，因此其是抗逆转录病毒治疗的主要靶标（例如在获得性免疫缺陷综合症(AIDS)中）。逆转录酶通常使用与各种引物复合的RNA作为DNA合成的模板。为了检测或定量RT活性，传统上通常对掺入的放射性标记核苷酸进行定量。
检测时间：大约4小时（对于1小时的酶反应来说），大约18小时（对于15小时的酶反应来说）。
样品材料：细胞培养上清液和其他生物样品、RT抑制剂。
灵敏度：逆转录酶检测可在1小时反应中检测到20pg HIV-1逆转录酶。可以在过夜反应中检测到1pg。
该检测方法可检测天然或重组逆转录病毒的逆转录酶活性，包括HIV-1、HIV-2、SIV-1、AMV和M-MulV的活性。
