检测原理: 欣博盛 QuantiCyto® ELISA试剂盒采用双抗体夹心法:抗人IL-2单抗包被于酶标板上,标本和标准品中的人IL-2会与单抗结合,游离的成分被洗去。加入生物素化的抗人IL-2抗体和辣根过氧化物酶标记的亲和素,生物素与亲和素特异性结合;抗人IL-2抗体与结合在单抗上的人IL-2结合而形成免疫复合物,游离的成分被洗去。加入显色底物,若反应孔中有人IL-2,辣根过氧化物酶会使无色的显色剂现蓝色,加终止液变黄。在450 nm处测OD值,人IL-2浓度与OD450值之间呈正比,可通过绘制标准曲线求出标本中人IL-2的浓度。 人IL-2简介: 白细胞介素2(IL-2)是T细胞和NK细胞产生的15.5kD糖蛋白,在机体的免疫应答中起重要作用。Morgan等人首先报道的名称是T细胞生长因子(T cell growth factor ,TCGF),1979年在第二届细胞因子研究会上定名为IL-2。IL-2的其它名称包括T细胞替代因子(T cell replacing factor,TRF)、T细胞促分裂因子(T cell mitogenic factor,TMF)淋巴细胞条件培养液(lymphocyte-conditioned medium,LCM)和杀伤辅助因子(killer helper factor,KHF)等。IL-2最主要的作用是诱导T淋巴细胞增殖(从G0期进入S期)和分化。人胸腺中的T细胞前体表达IL-2Rβ并分泌低水平的IL-2,这种自分泌作用可促进胸腺细胞增殖。IL-2参与调节T细胞受体基因的重排和表达。IL-2能诱导NK细胞增殖,增强NK细胞的活性,诱导LAK 细胞和TIL,并刺激它们产生细胞因子。高剂量IL-2能引起单核巨噬细胞增殖和分化,并增强单核巨噬细胞杀肿瘤细胞的作用。IL-2也能刺激已活化的B细胞增殖,活化B细胞表达高亲和力IL-2受体,其中a链由B细胞促分裂原诱导表达,而ß链由IL-2或IL-4诱导表达。IL-2除了支持B细胞生长外,还能促进免疫球蛋白(IgM和IgG)分泌,诱导J链合成从而加速IgM的装配和分泌。IL-2还刺激少突神经胶质细胞产生髓碱性蛋白,对该种细胞的增殖则呈双向性调节。在体内,IL-2有抗肿瘤、抗微生物感染、引起移植排斥和自身免疫以及免疫调节等作用。可溶性IL-2R作为免疫抑制剂,对白血病、移植排斥和自身免疫病可能有治疗作用。 部分文献引用: 1.
