    General description

    ββ-半乳糖苷酶或β-D-半乳糖苷半乳糖水解酶是由4条相同的各含1023个氨基酸的肽链组成的四聚体。对D-半乳糖表现高度专一性,完全激活需要K+或Na+和Mg2+。ββ-半乳糖苷酶催化含氧苷键的β-d-吡喃半乳糖苷的反应。

    37℃,以2-硝(xiāo)基苯基-β-D-半乳糖苷为底物,25℃,以乳糖为底物,约30 U/mg;用BSA标准化。

    Application

    ββ-半乳糖苷酶已用于酶联免疫吸附测定(ELISA)。使用本β-半乳糖苷酶可制作酶测定的校准曲线。

    切割单糖、寡糖或糖肽的1,4-连接末端半乳糖残基。

    Preparation Note

    硫酸铵制剂可以在2至8℃下稳定保存至标签注明的失效日期。可直接用于大多数应用,乳糖的定量。

    在不存在硫酸铵时,应使用Mg2+(89mM)和硫醇试剂(1mM β-巯基乙醇,1mM二硫苏糖醇)稳定β-半乳糖苷酶溶液[Beutler,1984]。硫醇可以减缓由分子内二硫键引起的酶二聚体形成。

    活化剂:需要K(50mM)进行激活(乳糖水解)。
    Na(50mM)也是一种活化剂,特别是对于2-硝(xiāo)基苯基-β-D-吡喃半乳糖苷的水解。
