磺丁基倍他环糊精钠的结构与组成
Cyclolab生产的磺丁基倍他环糊精钠是由β-环糊精与1,4-丁烷磺酸内酯发生取代反应的产物。β-CD葡萄糖单元是由7个吡喃葡萄糖通过α-1、4糖苷键连接而成,取代反应发生在β-CD葡萄糖单元的2,3,6碳羟基位置上,得到的产物是取代度为6.2~6.9的SBECD。
其结构如下图:

阴离子型高水溶性环糊精衍生物,能很好地与药物分子包合形成非共价复合物,从而提高药物的稳定性、水溶性、安全性,降低肾毒性、缓和药物溶血性,控制药物释放速率,掩盖不良气味等。
目前已应用于注射药、口服药、鼻部用药、眼部用药,对于含氮类药物具有特殊的亲和力和包合性。
磺丁基倍他环糊精钠在已上市制剂中的案例:
常用于难溶性药物,如伏立康唑,卡非佐米,齐拉西酮,马罗匹坦(动物药),阿立哌唑
泊沙康唑,卡马西平,美法仑,德拉沙星,甲苯咪唑,托吡酯,奥美拉唑,氯吡格雷,多西他赛,美洛昔康,四氢孕酮以及其他几种含氮的API碱处于不同的临床阶段

应用案例:
注射用瑞德西韦(Remdesivir)已经在中日友好医院开始抗新冠状病毒(2019-nCoV)的三期临床研究。此前吉利德公司已经对瑞德西韦(Remdesivir)的剂量和安全性做了不少研究。在临床剂量下,瑞德西韦(Remdesivir)除对肝功能有影响外,表现出很好的总体安全性。
瑞德西韦冻干制剂还包含以下非活性成分:注射用水,磺丁基-β-环糊精钠(SBECD), 使用HCL和/或NAOH将制剂的pH调节至3.0至4.0,而后冻干。按照目前的稳定性数据,该冻干剂可以在30度以下保存三年。
