  本酶是通过蛋白质工程改造的phi29 DNA 聚合酶在大肠杆菌中进行表达后，经多次纯化分离获得。与天然phi29 DNA 聚合酶相比，具有更高的扩增效率和灵敏度。Phi29DNA聚合酶具有高效的DNA合成能力，可连续合成多达70kb的DNA片段。同时该酶具有3´→5´外切酶校读功能，保真性高，还具有特殊的链置换和连续合成特性。

图例一）Super phi29 DNA Pol扩增
以菌液为模板，扩增pUC19质粒，自左向右模板量分别为10μl反应体系中加入0.5μl，1μl，2μl，3μl菌液做模板。左图：扩增产物直接电泳，由于产物量较大，部分DNA未电泳出加样孔（上样量2μl）；右图：扩增产物EcoRI酶切后电泳，显示与pUC19大小一致的均一条带。

1，2，3与4，5，6分别为扩增1小时，2小时，3小时的结果。10μl反应体系中加入1ng人基因组DNA，上样量2μl。1-3：野生型酶；4-6：Super phi29 DNA Polymerase。

10×phi29 Buffer	5μl
随机引物（1mM）	1.25μl

加ddH2O至45μl,95℃预变性3min,冰上冷却。

质量保证：经多次柱纯化，SDS-PAGE胶检测仅可见清晰单一的目的条带；PCR方法检测无大肠杆菌DNA残留，无核酸内、外切酶污染。

活性定义：在30℃条件下，10min内能使0.5 pmol的dNTP掺入酸不溶性物所需要的酶量定义为1个活性单位。

10×反应缓冲液：500mM Tris-HCl（pH 7.5）, 100mM MgCl2，100mM (NH4)2SO4，40mM DTT。反应时需要加入dNTP。

关于CDH11：
Cadherin-11, also known as OSF-4, Osteoblast cadherin and CDH11, is a type II classical cadherin from the cadherin superfamily, integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Cadherins interact with themselves in a homophilic manner in connecting cells, may thus contribute to the sorting of heterogeneous cell types. Cadherin-11 contains five cadherin domains and is mainly expressed in brain. Mature cadherin proteins consists of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small, highly conserved C-terminal cytoplasmic domain. It is shown that cadherin-11 is a viable molecular target for therapeutic intervention in Glioblastoma multiforme.

Hemoglobin subunit theta-1 is a protein that in humans is encoded by the HBQ1 gene. Theta-globin mRNA is originally found in human fetal erythroid tissue but not in adult erythroid or other nonerythroid tissue. Theta-1 is a member of the human alpha-globin gene cluster that includes five functional genes and two pseudogenes. Research supports a transcriptionally active role for the gene and a functional role for the peptide in specific cells, possibly those of early erythroid tissue. Hemoglobin has a quaternary structure characteristically composed of many multi-subunit globular proteins. Most of the amino acids in hemoglobin form alpha helices, connected by short non-helical segments. Hydrogen bonds stabilize the helical sections inside this protein, causing attractions within the molecule, folding each polypeptide chain into a specific shape. Hemoglobins quaternary structure comes from its four subunits in roughly a tetrahedral arrangement.

Cyclophilins(CyPs) are a family of proteins found in organisms ranging from prokaryotes to humans. These molecules exhibit peptidyl-prolyl isomerase activity, suggesting that they influence the conformation of proteins in cells. Cyclophilin A accelerate the folding of proteins. It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. Cyclophilin A can interact with several HIV proteins, including p55 gag, Vpr, and capsid protein, and has been shown to be necessary for the formation of infectious HIV virions.
