关于DNAM-1/CD226：
Mouse DNAX accessory molecule-1(DNAM-1) is a type I transmembrane glycoprotein that belongs to the immunoglobulin superfamily. As an activating receptor, it interacts with the ligands CD155 and CD112, and activates natural killer (NK) cells via its immu-noreceptor tyrosine-based activatory motif (ITAM). Mature mouse DNAM-1 has extracellular domain (ECD) that contains two Ig-like C2-set domains, and possesses a cytoplasmic region that contains motifs for binding PDZ domains. DNAM-1 is expressed on several lymphoid and myeloid cell types and interacts with CD155/PVR and Nectin-2/CD112. Ligation of DNAM-1 promotes the activation of NK cells, CD8+ T cells, and mast cells, induces dendritic cell maturation, initiates megakaryocyte and activated platelet adhesion to vascular endothelial cells, and stimulates monocyte extravasation; Conversely, it inhibits the formation of osteoclasts. Platelet-endothelium interactions that are mediated by DNAM-1 enable the metastasis of tumor cells to the lung.

关于BLNK：
B-Cell Linker Protein (BLNK) is a cell membrane protein which contains 1 SH2 domain. BLNK is expressed in B cells and fibroblast cell lines, playing a important role in B cell receptor signaling. BLNK as a central linker protein, downstream of the B-cell receptor (BCR), bridges the SYK kinase to a multitude of signaling pathways and regulating biological outcomes of B-cell function and development. BLNK associates with the activation of ERK/EPHB2, MAP kinase p38 and JNK, modulates AP1, NF-kappa-B and NFAT activation. BLNK involves in BCR-mediated PLCG1 and PLCG2 activation and Ca2+ mobilization and is required for trafficking of the BCR to late endosomes. BLNK deficiency results in agammaglobulinemia type 4 and much more profound block in B-cell development.

CD48 antigen, also known as B-lymphocyte activation marker BLAST-1, BCM1 surface antigen, Leukocyte antigen MEM-102, TCT.1, CD48, BCM1,and BLAST1, CD48 contains one Ig-like C2-type domain and one Ig-like V-type domain, but does not have a transmembrane domain, however, but is held at the cell surface by a GPI anchor via a C-terminal domain which maybe cleaved to yield a soluble form of the receptor. CD48 may facilitate interaction between activated lymphocytes and be involved in regulating T-cell activation. CD48 plays a vital role as an environmental sensor for regulating progenitor cell numbers and inhibiting tumor development. It is suggested that the anti-CD48 mAb has the potential to become an effective therapeutic mAb against multiple myeloma.
