72 kDa type IV collagenase also known as matrix metalloproteinase-2 (MMP-2) and gelatinase A is an enzyme that in humans is encoded by the MMP2 gene.It belongs to the matrix metalloproteinase (MMP) family. Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that degrade components of the extracellular matrix (ECM) and play essential roles in various physiological processes such as morphogenesis, differentiation, angiogenesis and tissue remodeling, as well as pathological processes including inflammation, arthritis, cardiovascular diseases, pulmonary diseases and tumor invasion. MMP-2 is ubiquitinous metalloproteinase that is involved in diverse functions such as remodeling of the vasculature, angiogenesis, tissue repair, tumor invasion, inflammation, atherosclerotic plaque rupture, as well as degrading extracellular matrix proteins. MMP-2 can also act on several nonmatrix proteins such as big endothelial 1 and beta-type CGRP promoting vasoconstriction.   本制品是通过基因重组技术克隆表达的缺失突变型RNase H-的M-MuLV反转录酶。一般的野生型M-MuLV（Moloney Murine Leukemia Virus）具有以下几种活性：依赖于RNA的DNA聚合酶活性；依赖于DNA的DNA聚合酶活性；RNase H活性。由于RNase H能够催化降解DNA/RNA杂合体中的RNA，因此在cDNA第一条链的合成反应中可能会降解RNA/DNA杂合体中的模板RNA。本酶的RNase H活性缺失，延伸能力强，可用于较长的cDNA合成，高比例的全长cDNA文库的构建以及Real Time RT-PCR反应等。

1、1st-Strand cDNA的合成。
2、cDNA Probe的制备。

注意事项：
1、用DEPC处理实验用到的所有管子和枪头，或者购买已经证明无核酸酶的实验用具，整个实验过程戴手套并经常更换手套，避免RNA酶的污染。
2、确保所用试剂及超纯水中无RNA酶污染。
3、纯化过的RNA要保证不含有盐，金属离子，乙醇和*酚，因为以上成分会干扰第一链cDNA的合成反应。可采用乙醇沉淀RNA的方法去除掉痕量污染物。

实验步骤：
第一链cDNA合成反应体系：
1、按右侧表格顺序加入的反应物。
2、可选优化步骤。如果RNA模板GC含量高或者含有二级结构，可先将模板和引物的 混合液轻轻混匀，短暂离心，65℃孵育5分钟，冰上冷却，离心，再置于冰上冷却。
3、轻轻混匀，离心。
4、如果使用Oligo(dT)18或者基因特异型引物，42℃孵育60分钟。如果使用随机六聚体引物，25℃先孵育5分钟，随后42℃孵育60分钟。
5、75℃加热5 min终止反应。
反应产物可直接用于PCR反应或者在-20℃保存少于一周的时间。如想延长保存时间，建议使用-70℃保存。

应用实例：

图例一）小鼠胚肝总RNA RT-PCR扩增。
泳道1-6总RNA量分别为10ng，1ng，0.1ng，0.01ng，0.001ng，0ng；20ul体系中反转录后取1ul用于PCR扩增。目的基因：GAPDH。

质量保证：经多次柱纯化，SDS-PAGE检测纯度大于95%，PCR方法检测无大肠杆菌DNA残留，无核酸内、外切酶污染。

活性定义：以Poly（rA）•Oligo（dT）为模板/引物，在37℃、10分钟条件下，掺入1 nmol的[3H] dTTP所需要的酶量定义为1个活性单位（U）。
