产品描述
HNF4荧光素酶报告基因（报告基因质粒）（HNF4 luciferase reporter plasimd）是翊圣生物自主研发的用于检测HNF4转录活性水平为目的的报告基因。HNF4(Hepatocyte Nuclear Factor 4)是转录因子核受体超家族中的一员，参与肝细胞分化调控和维持肝细胞的生物学功能。HNF4α主要在消化系统、肝脏和肾脏表达，以及在白色脂肪组织和胰腺中也有少量的表达。HNF4α具有阻断肝纤维化、肝硬化、肝癌的疾病进程,改善肝脏功能的作用。另外,HNF4α在肝干细胞移植方面也发挥重要作用,提高肝细胞移植的成功率。
HNF4报告基因主要用于检测细胞中HNF4的转录活性、药物研究以及基因过表达和RNAi的表型分析等。
pGMHNF4-Lu是翊圣生物改造后的哺乳动物真核表达载体，在其多克隆位点插入了多个HNF4结合位点，可以高灵敏度地检测HNF4的激活水平。同时，对载体中预测出的其它转录因子以外的结合位点进行了适当的突变，增加了质粒的转录因子结合特异性。由于质粒体积减小，使得HNF4报告基因质粒更易于转染。
