DNA 聚合酶 I（ E.coli ）是依赖于 DNA 的 DNA 聚合酶，具有 3´ → 5´ 和 5´ → 3´ 的外切核酸酶的活性。该酶的 5´ → 3´核酸外切酶活性能够从新生成的DNA 链的5´ 端切除核苷酸，从而使切刻平移成为可能。该酶是一种重组 E.coli 菌株，携带有高表达的 polA 基因。
用途
• DNA 切刻平移，制备具有高比活性的探针
• cDNA 第二条链的合成

活性定义（粘性末端活性单位）
1 单位指在37℃ 反应条件下， 30 分钟能使 10 nmol 的dNTPs 掺入酸不溶性沉淀物所需要的酶量定义为1 个活性单位。
热失活
75℃ 温育20 分钟。

该酶不含脱氧核糖核酸酶I，因此用于切刻平移反应时需要另加入脱氧核糖核酸酶I。
