Single Cell Full Length mRNA-Amplification Kit
第一链cDNA合成， 全长cDNA扩增，产物可用于基因表达差异；融合表达，可变剪切分析
模板起始量低：单个细胞或10pg Total RNA即可作为起始模板，进行高效扩增
扩增灵敏度高：使用LNA技术及精心优化的反应体系，提高低表达基因的检出量
产物完整性好：通过双端引物扩增全长cDNA，获得全转录组信息，避免了5'和3'偏好性
单个293T细胞和10 pg Total RNA分别扩增18个循环，取1 µl纯化后的cDNA扩增产物使用Agilent 2100 Bioanalyzer进行检测，扩增产物分布于400-10000 bp，文库的peak位于2000 bp 左右；无模板阴性对照无产物。
Single Cell Full Length mRNA Amplification Kit能以1-500个细胞或10 pg-10 ng总RNA为模板，以Oligo(dT) VN Primer 为逆转录引物进行cDNA的合成，并且利用逆转录酶Sc Reverse Transcriptase的Template-switching活性在cDNA的3’端添加一段接头序列，通过该接头序列进行后续PCR扩增以获得全长cDNA扩增产物。有效避免了cDNA合成过程中的3’偏好性和rRNA的污染,对获得的全长cDNA扩增产物进行测序分析可以得到基因表达差异、可变剪接、融合基因等遗传调控信息。
Single Cell Full Length mRNA Amplification Kit ，兼容样品体积可控制为2.5 μl。一般情况下，一个反应可以产生2-20 ng的cDNA扩增产物。
