E. coli BL21(DE3)是T7或T7/lac表达系统常用的表达菌株，其基因型为F- lon ompT hsdSB (rB-,mB-) dcm gal rne131(DE3)。T7 RNA聚合酶基因的表达受控于l噬菌体DE3区的lacUV5启动子，该区整合于BL21的染色体上。该菌适合于非毒性蛋白的重组表达。使用pUC19质粒检测，转化效率可达106 cfu/μg DNA。
