Background	Integrins are transmembrane receptors that mediate the attachment between a cell and its surroundings, such as other cells or the extracellular matrix (ECM). Integrins are obligate heterodimers containing two distinct chains, called the α (alpha) and β (beta) subunits. The molecular mass of the integrin subunits can vary from 90 kDa to 160 kDa. Beta subunits have four cysteine-rich repeated sequences. Both α and β subunits bind several divalent cations. Integrins have two main functions: Attachment of the cell to the ECM and signal transduction from the ECM to the cell. However, they are also involved in a wide range of other biological activities, including immune patrolling, cell migration, and binding to cells by certain viruses, such as adenovirus, echovirus, hantavirus, and foot and mouth disease viruses. Research studies have implicated β1 integrin in various activities including embryonic development, blood vessel, skin, bone, and muscle formation, as well as tumor metastasis and angiogenesis.

Background	β-Catenin is a key downstream effector in the Wnt signaling pathway. It is implicated in two major biological processes in vertebrates: early embryonic development and tumorigenesis. Phosphorylation at Ser-552 by AMPK promotes stabilizion of the protein, enhancing TCF/LEF-mediated transcription. Phosphorylation on Tyr-654 decreases CDH1 binding and enhances TBP binding. GSK-3β destabilizes β-catenin by phosphorylating it at Ser33, Ser37, and Thr41. Mutations at these sites result in the stabilization of β-catenin protein levels and have been found in many tumor cell lines. In the presence of Wnt ligand, CTNNB1 is not ubiquitinated and accumulates in the nucleus, where it acts as a coactivator for transcription factors of the TCF/LEF family, leading to activate Wnt responsive genes.

本探针是用于EMSA(也称gel shift)研究的β-Catenin/TCF consensus oligonucleotide的突变体。可以作为EMSA探针－β-Catenin/TCF的阴性对照，用于EMSA结合反应中突变探针的冷竞争反应等。 一个包装的突变探针，如果用于同位素标记EMSA探针的突变探针冷竞争反应，可以进行90-180个突变探针的冷竞争反应。如果用于生物素标记探针的冷竞争时，可以进行约30个冷竞争反应。

β-Catenin/TCF consensus oligo的序列如下：
5"-CCC TTT GAT CTT ACC-3"
3"-GGG AAA CTA GAA TGG-5"

EMSA突变探针－β-Catenin/TCF中β-Catenin/TCF的公认的结合位点发生了突变，从而使β-Catenin/TCF无法和该突变探针结合。在探针冷竞争反应中，正常的标记探针和β-Catenin/TCF的结合的条带会被抑制；而在突变探针冷竞争反应(cold competition)中，正常的标记探针和β-Catenin/TCF的结合的条带不会被抑制。参考下图。


一个典型的EMSA/Gel- Shift分析图
1,阴性对照反应(标记探针)；
2,常规反应(含激活的目的转录因子的核蛋白＋标记探针)；
3,探针冷竞争反应(含激活的目的转录因子的核蛋白＋标记探针＋标记探针100倍量的未标记探针)；
4,突变探针的冷竞争反应(含激活的目的转录因子的核蛋白＋标记探针＋标记探针100倍量的未标记突变探针)；
5,Super-shift反应(含激活的目的转录因子的核蛋白＋标记探针＋目的转录因子的特异抗体)。

注意事项：
1. 避免加热到40℃以上，温度过高会导致双链DNA探针解聚成单链。而单链无法用于EMSA研究。
