柑橘叶片植物标本的采集及保存
1、 新鲜植物组织请在液氮中充分研磨；
2、 加入样品体积3倍的提取液（10% TCA）, 置于-20 过夜；
3、 请于4 ，8000rpm，离心1小时，弃上清，收集沉淀；
4、 沉淀加入等体积的冰浴丙酮，混匀后于4 离心（8000rpm，15分钟），弃上清并真空干燥，保存备用；
5、 上样前加入裂解液（2.7g 尿素，0.2g CHAPS，溶于去离子水中至终体积5ml），混匀后室温放置30分钟，然后4 离心（8000rpm，15分钟），取上清并暂时保存于4 待用。
关于样本：
我们在收集产品标本前都必须有一个完整的计划，必须清楚要检测的成份是否足够稳定。我们提倡新鲜标本尽早检测,
对收集后当天就进行检测的标本，及时储存在4℃备用，如有特殊原因需要周期收集标本，请造模取材后，将标本及时分装后放在-20℃或-80℃条件下保存。因冰室与室温存在一定温差，蛋白极易降解，直接影响实验质量，所以避免反复冻融.
样本要求
1、样本不能含叠氮钠（NaN3），因为叠氮钠（NaN3）是辣根过氧化物酶（HRP）的抑制剂。
2、标本采集后尽早进行提取，提取按相关文献进行，提取后应尽快进行实验。若不能立即试验，可将标本放于-20℃保存，但应避免反复冻融。
3、样本应充分离心，不得有溶血及颗粒。
