Monoclonal Antibody against Phosphorylated Neurofilaments H and M Intended Use **Research Use Only (RUO)**
This product is sold for laboratory research use only, not for human or in-vivo use.Clone SMI-310 Form Ascites Fluid (contains 0.01M sodium azide) Host Mouse Species Reactivity Mammalian IsoType Neurofilament H Phosphorylated (SMI 310) Monoclonal AntibodyIgG1 Specificity SMI 310 reacts with a phosphorylated epitope in extensively phosphorylated neurofilament H and, to a much lesser extent, with neurofilament M in most mammalian species. Immunocytochemically, SMI 310 reacts with selected axons. The extent of reaction with axons is more restricted than that of any other antiphosphoneurofilaments. Cell bodies and dendrites are generally unstained. Other cells and tissues are unreactive except for peripheral axons. Phosphatase treatment of tissue sections or Western blots abolishes reaction with SMI 310. SMI 310 needs less phosphatase treatment of neurofilaments for abolition of reactivity than any other antibody to phosphorylated neurofilaments, suggesting that a very extensive degree of hyperphosphorylation of neurofilaments is necessary for its reactivity. In pathological conditions, reaction with SMI 310 may be found also in neuronal cell bodies. In Alzheimers disease, SMI 310 provides a strong reaction with extraneuronal (ghost) neurofibrillary tangles. Intraneuronal tangles remain unreactive. SMI 310 may react with an unusual and sparsely distributed form of phosphorylated neurofilaments that may be exacerbated in certain pathologic conditions. SMI 310 distinguishes sporadic inclusion-body myositis from hereditary inclusion-body myopathies. Uses This antibody is effective in immunoblotting, immunocytochemistry and ELISA. Suggested Working Dilution Neurofilament H Phosphorylated (SMI 310) Monoclonal AntibodyThe extent of permissible dilution of SMI 310 beyond those recommended for general application depends upon nature and concentration of the antigen examined, species of the antigen, method of fixation and kind of section examined. - Western blot: 1:1,000

Tissue Preparation: SMI 310 reacts on Western blots, paraffin, vibratome and frozen tissue sections as well as with neuronal cell cultures. Tissues and cultures can be fixed in a variety of paraformaldehyde or formaldehyde-containing fixatives including Bouins. "Recovery" of antigen in formalin-fixed tissue can be accomplished by autoclaving of deparaffinized sections in distilled water. Post-fixation in cold methanol or methanol/hydrogen peroxide facilitates access of SMI 310 in frozen sections or thick sections of tissue perfused with 4% paraformaldehyde and in tissue cultures. In tissue that has not been paraffin embedded, enhanced reaction for neurofilaments and other antigens in material that has been stored for a long time in formaldehyde fixatives can be obtained by boiling 0.5 cm thick tissue blocks immersed in Tris buffered saline, pH 9.0. in a microwave oven for 15 min prior to sectioning. These antibodies can be used for neurofilament detection in trypsin-treated tissue.Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C.
