Assay procedure summary
1. Prepare all reagents, samples and standards;
2. Add 100µL standard or sample to each well. Incubate 2 hours at 37oC;
3. Aspirate and add 100µL prepared Detection Reagent A. Incubate 1 hour at 37oC;
4. Aspirate and wash 3 times;
5. Add 100µL prepared Detection Reagent B. Incubate 30 minutes at 37oC;
6. Aspirate and wash 5 times;
7. Add 90µL Substrate Solution. Incubate 15-25 minutes at 37oC;
8. Add 50µL Stop Solution. Read at 450nm immediately.

Test principle
β-血小板球蛋白(βTG)检测试剂盒The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Beta-Thromboglobulin (bTG). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Beta-Thromboglobulin (bTG). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Beta-Thromboglobulin (bTG), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Beta-Thromboglobulin (bTG) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

含于乳中的球蛋白类的一种蛋白质。牛乳中所含全蛋白（3.7％）的 80％为酪蛋白，在除去酪蛋白的乳清蛋白质中 55％为β -乳蛋白。可获得其结晶，等电点 =5.2，在此 pH下主要成双量体结构。单量的分子量为 1万 8千，已知由遗传所决定的有 A、 B、 C三种分子类型。从一头牛所得的样品中仅由一种或两种等量的混合物组成。三种类型分子的不同，在于一个分子中有两个或一个氨基酸残基被进行置换（ A的 ASP， Val分别对 Gly和 Ala， B的 Gln，在 C对 His）。单一的β
