Stemolecule™ Wnt Inhibitor IWP-3 was identified in a high throughput screen for antagonists of the Wnt/β-catenin pathway1. Wnt Inhibitor IWP-3 prevents palmitylation of Wnt proteins by Porcupine (Porcn), a membrane-bound O-acyltransferase, thereby blocking Wnt secretion and activity1. It has an IC50 of 40 nM and blocks phosphorylation of the Lrp6 receptor and accumulation of both Dvl2 and β-catenin1.
Soluble in DMSO (100 mM). For a 10 mM concentrated stock solution, reconstitute the compound by adding 413 µl of DMSO to the entire contents of the vial. Note: for most cells, the maximum tolerance to DMSO is <0.5%.

Store at 4°C. Protect from light. Following reconstitution, store aliquots at -20°C.
Stability
Stock solutions stable at -20°C for 6 months.
Quality control
The purity of Wnt Inhibitor IWP-3 was determined by HPLC analysis. The accurate mass and structure of Wnt Inhibitor IWP-3 were determined by mass spectrometry and NMR, respectively. Cellular toxicity of Wnt Inhibitor IWP-3 was tested on mouse ES cells.
