CD74 / DHLAG / p33 抗体背景综述
Human CD74, also known as HLA-DR antigens-associated invariant chain or Ii, is a non-polymorphic glycoprotein, which is expressed on antigen-presenting cells (APC), and carcinomas of renal, lung, gastric, thymic origin and by certain sarcomas. It is post-translationally glycosylated and exists in different isoforms. The most common isoform is 33 kDa (p33), but p35, p41 and p43 isoforms also exist. The most well-known function of CD74 is regulating the trafficking of class Ⅱ major histocompatibility complex (MHC) proteins in APC. It also acts as a surface membrane receptor for macrophage migration inhibitory factor (MIF), alone or together with CD44. But multiple studies have revealed that cell surface expression of CD74 is not always dependent on class Ⅱ MHC. The role of CD74 in signal transduction was initially hypothesized when it was found to be phosphorylated and associated with proteins that coordinate various signal transduction pathways. CD74 was recently found to be increased in microglia in Alzheimer disease (AD) cases compared to age-matched controls. Additionally, it may be useful targets to reduce neutrophilic lung inflammation, and acute lung injury. Various studies have indicated that CD74 is highly expressed in inflammatory disorders and cancers. Thus, the restricted expression of CD74 by normal tissues and its very rapid internalization make CD74 an attractive therapeutic target for both cancer and immunologic diseases.
