FKBP1A / FKBP12抗体背景综述
FK506 binding protein, 12 kilodalton molecular weight (FKBP12), also known as FKBP1, along with cyclophilin, are two major members of the immunophilin protein family who serve as receptors for the immunosuppressant drugs cyclosporin A and FK506. As a conserved molecules in many eukaryotes, FKBP12 has been characterized as a peptidyl-prolyl isomerase that catalyzes the transition between cis- and trans-proline residues, and is involved in several biochemical processes including protein folding, receptor signaling, protein trafficking and transcription. FK506 which is used in treating patients after organ transplant and patients suffering from autoimmune disorders is inactive by itself and requires binding to FKBP-12 or other immunophilins for activation.This FK506-FKBP12 complex inhibits the activity of the serine/threonine protein phosphatase 2B, calcineurin, thus blocking signal transduction in the T-lymphocyte transduction pathway. However, FKBP12 is concentrated more than 10 times higher in the brain than in immune tissues, the decreased activity of calcineurin will increase the phosphorylated levels of its substrates, and thus results in the degenerative diseases of CNS, such as Parkinson and Alzheimers Disease.
