IL4R / IL4Ra / CD124抗体 Background
IL-4 is a pleiotropic cytokine produced by activated Th2 cells and mast cells, and plays a pivotal role in immune responses. The effects of IL-4 are mediated after binding to high affinity receptor complexes present on hematopoietic as well as non-hematopoietic cells. Hematopoietic cellular responses to IL-4 are mediated by a high affinity receptor complex comprised of the IL4Rα (CD124) subunit and the common cytokine γc chain (CD132). In non-hematopoietic cells, the IL4Rα subunit associates with the IL13Rα chain and also serves as a functional complex for IL-13. IL4Rα is a single-pass type I transmembrane protein containing ser/Pro-rich regions in cytoplasmic domain similar to those present in the IL-2 receptor and GM-CSF receptor ß-chains. As a receptor for both IL4 and IL13, IL4Rα couples to the JAK1/2/3-STAT6 pathway, and is involved in regulating IgE production and, chemokine and mucus production at sites of allergic inflammmation, as well as promoting differentiation of Th2 cells. Two transcript variants encoding different isoforms of IL4Rα, a membrane-bound and a soluble form, have been found, and the soluble form of IL4Rα can inhibit IL4-mediated cell proliferation and IL5 upregulation by T-cells.
