MSR1 / SCARA1 / CD204抗体背景综述
Scavenger receptors are a group of membrane receptors that recognize and internalize modified low density lipoproteins (LDLs) and participate in the removal of many foreign substances and waste materials in the living body. Scavenger receptors are categorized into three classes, and class A mainly expressed in macrophage includes three isoforms derived by alternative splicing. The class A macrophage scavenger receptor are trimers with a molecular weight of about 220-250 kDa, and isoformⅠ( SR-AI or MSR-AI ) contains six predicted structural domains: cytoplasmic, transmembrane, spacer, α-helical coiled coil, collagenous, and an isoform-specific cysteine-rich C-terminus which are truncated in SR-AII and SR-AIII. SR-AI /II are functional receptors and are able to mediate the endocytosis of low density lipoproteins (LDLs) modified by oxidation or acetylation., nucleic acids, and a variety of plant and microbial cell wall constituents, whereas SR-A III is trapped in the endoplasmic reticulum and acts as a dominant negative protein with respect to SR-A activity. Accordingly, SR-A has been implicated in the development of atherosclerotic lesions and other macrophage-associated host defense reponses. Also, SR-A has been suggested to play important roles in macrophage growth, cell adhesion, osteoclast differentiation, and as well as intracellular signaling.

因为抗体的结合而减少;在人体血清中无游离的CD20存在;最重要的是B淋巴细胞瘤上的CD20少有内吞和脱落的发生[ 2-5 ] 。1997年,以CD20为靶点的嵌合抗CD20单克隆抗体Rituxan已获准应用于B细胞淋巴瘤的治疗[ 6 ]。HI47(IgG3)是我所于1990年研制成功的国内第一个抗CD20鼠源性单克窿抗体,第四届国际人类白细胞分化抗原会议将HI47命名为CD20+X[ 7 ] 。我们利用噬菌体显示技术从HI47杂交瘤细胞中克隆到抗CD20抗体HI47可变区基因,将该基因构建成为嵌合的抗
20B淋巴细胞表面的膜蛋白CD20对B淋巴细胞的增殖和分化具有调节作用[1]。由于CD20分子仅在前-B淋巴细胞、未成熟B淋巴细胞、成熟B淋巴细胞、激活B淋巴细胞中表达,而在浆细胞、淋巴多能干细胞以及其它组织均无CD20的表达[2,3,4],在人体血清中亦无游离CD20的存在[5],因此CD20可作为B淋巴细胞瘤治疗的一个理想治疗靶点。限制抗体临床使用的因素主要有抗体产生的免疫原性和抗体的生产能力是否能够满足临床需要。第四届国际人类白细胞分化抗原会议将鼠源性单克隆抗CD20抗体HI47(IgG3)正式命名为CD20+X
