一般描述
焦碳酸二乙酯（DEPC）是一种用于灭活RNase酶并对水分和pH敏感的化学制品。在155℃时它在水溶液中可分解成乙醇和二氧化碳。DEPC对能将其分解成氨基甲 酸乙酯这种潜在致癌物的氨也很敏感。
应用
焦碳酸二乙酯已在PCR反应中用于处理去离子水，以降低了RNA被RNase降解的风险。[1]它还可以用于斑点印迹杂交，以稀释分离自不同微生物的总RNA。[2]
蛋白中His及Tyr残基的修饰试剂。用于dsDNA结构破坏的稳健探针，可与完全或部分未堆叠的碱基发生反应。
生化/生理作用
DEPC是一种潜在的核酸酶抑制剂。[3][4]0.1%的DEPC溶液用于灭活RNase。
它可以剂量和时间依赖的方式抑制兰尼碱与骨骼肌中兰尼碱/ Ca2+受体通道的结合，并提高SR囊泡的Ca2+通透性。[5]
在溶液中浓度约为 0.1% (v/v) 时可使 RNase 失活，从而防止 RNA 被降解。
