Biological Activity
GW5074 is a potent, selective and cell-permeable inhibitor of c-Raf (IC50 = 9 nM) with no effect on the activities of cdk1, cdk2, c-src, p38 MAPK, VEGFR2 and c-fms [1]. GW5074 blocks apoptosis in cerebellar granule neurons induced by diverse stimuli such as LK-treatment or exposure to MPP+ or methylmercury, and also prevents oxidative stress-induced apoptosis in cortical neurons. GW5074 prevents neurodegeneration and improves behavioral outcome in an animal model of Huntington’s disease [2]. GW5074 attenuates VEGF-induced hyperpermeability [3] and significantly suppresses the airway hyperreactivity in a mouse model of sidestream smoke exposure [4].
