一般描述:蟾蜍灵(Bufalin)是一种C-24类固醇,在C-17处带有一个α-吡喃酮环。它存在于蟾蜍(Venenum Bufonis)的毒液腺和皮肤中。蟾蜍灵是强心类固醇,是蟾蜍的地(dì)(gāo)辛类成分。
应用,蟾蜍灵已被用于:
- 测试其对核因子κB(NF-κB)细胞和肿瘤坏死因子-α(TNF-α)刺激的类风湿关节炎成纤维样滑膜细胞(RAFLS)的侵袭和激活的影响
- 作为软骨细胞中的核受体共激活因子3(NCOA3)抑制剂
- 用于测试其在对吉非替尼有抗药性的肺癌细胞(NCI-H460)中的抗转移作用
生化/生理作用
蟾蜍灵是一种抗转移药,通过促进包括肝细胞癌、膀胱癌和骨肉瘤在内的各种癌症的细胞周期阻滞(cell cycle arrest )和细胞凋亡来引发抗肿瘤功能。它还促进自噬细胞死亡并具有抗炎特性。蟾蜍灵是一种类固醇受体共激活剂(SRC)抑制剂。
