描述:在实体组织和细胞内,胆固醇酯既是构成细胞质膜的成分,也是细胞内脂滴的主要组分。细胞内胆固醇过度聚集与动脉粥样硬化泡沫细胞形成密切相关。实体组织和细胞的胆固醇测定远比血液胆固醇测定复杂。本试剂盒避免了有毒的有机溶剂抽提、繁杂的氮气吹干和脂质复溶等步骤,采用高效能试剂裂解细胞和提取胆固醇,优化了酶学反应和操作步骤,简单易行,灵敏度高,线性范围20~5000 µmol/L。
原理:本试剂盒采用WHO、中国《全国临床检验操作规程》推荐的酶学方法,结合经典GPO Trinder酶学反应1,2,原理如下:(1) 胆固醇酯酶分解胆固醇酯为游离胆固醇。(2) 胆固醇氧化酶将游离胆固醇氧化,反应过程产生过氧化氢。(3) 在过氧化物酶催化下进行生色反应,在550nm有最大吸收峰,光密度值与胆固醇浓度成正比。
