跨膜糖蛋白组织因子（TF）是生理性凝血过程的启动物，TF可结合并激活VII，TF-VIIa复合物又可进一步激活因子IX和因子X，导致凝血酶的生成。很多病理条件下，如革兰氏阴性败血症、急性冠脉综合症等都可诱导TF的表达，后者与这些疾病过程中危及生命的血栓形成有关，败血症中血管内TF表达可导致弥散性血管内凝血。TF还在血管发生、转移、及肿瘤相关的血管生成方面发挥重要作用。
Principle of the Assay
The AssayMax Human Tissue Factor (TF) ELISA kit is designed for detection of human TF in plasma, serum, tissue extracts, and cell culture samples. This assay employs a quantitative sandwich enzyme immunoassay technique that measures TF in less than 5 hours. A polyclonal antibody specific for TF has been pre-coated onto a 96-well microplate. TF in standards and samples is sandwiched by the immobilized antibody and the biotinylated polyclonal antibody specific for TF, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
