小鼠全血：包含红细胞（无核）、白细胞、血小板、血浆。小鼠血液离体凝固极快，采血必须立刻与抗凝剂轻柔混匀；不抗凝静置凝固后离心得到血清；抗凝后离心得到血浆。

小鼠总血量≈6‑8 mL /100 g 体重，20 g 小鼠总血量约 1.2‑1.6 mL。

常用采血方式

1. 眼眶静脉丛（非终末）：麻醉，毛细管刺入内眼角，采血量 200‑500 μL，可多次采血。
2. 摘眼球（终末）：血量 0.6‑1.0 mL，血液靠重力流出，禁止挤压胸腔，避免溶血、混入组织液。
3. 心脏穿刺（终末，血量最大）：麻醉仰卧，3‑4 肋间进针，可采集 0.8‑1.2 mL，适合无菌大量样本吉林大学第...。
4. 尾尖采血：仅几十 μL，适合微量检测。

抗凝剂选择（重点，不同实验不能混用）

操作：采血前把抗凝剂加到 EP 管，采血滴入后轻柔颠倒 5‑8 次混匀，严禁剧烈震荡，防止溶血。

血浆制备（抗凝全血）

1. 抗凝全血，4℃，1000‑1500 g，离心 10‑15 min
2. 小心吸取上层淡黄色上清即为血浆，避免吸到白细胞层
3. 分装，短期 4℃；长期‑80℃，禁止反复冻融

血清制备（不加抗凝剂）

1. 血液收集无抗凝 EP 管，室温静置 30‑60 min 充分凝固
2. 4℃ 1500‑2000 g 离心 10‑15 min，吸取上层黄色上清，即为血清。

小鼠红细胞裂解（全血提白细胞，流式常用）

1. 取抗凝全血，加入红细胞裂解液（ACK 裂解液）
2. 室温避光裂解 3‑5 min，液体透亮红色
3. PBS 终止裂解，离心弃红色上清，沉淀即为白细胞。

关键注意事项

1. 小鼠血液极易凝固，采血与混匀动作要快，出现凝块样本直接报废。
2. 全程避免溶血：不要挤压组织、不要剧烈震荡，离心转速不要过高。
3. 终末采血需要遵循实验动物伦理规范。
4. 样本尽量采血后 30min 内完成处理。

对比记忆：鸡血红细胞有核，小鼠红细胞无核，所以巨噬细胞吞噬实验用鸡血红细胞，不用小鼠红细胞。
