Detection Principle:
The Human Amylase ELISA Kit uses a sandwich ELISA format. The plate is pre-coated with anti-amylase capture antibody. Amylase in the sample binds to the immobilized antibody, followed by HRP-conjugated detection antibody. After TMB substrate addition, color intensity is directly proportional to the amylase concentration.
Operating Steps:

| 3 | Incubate at 37°C for 2 hours |
| 4 | Wash 3 times with wash buffer |
| 5 | Add 100μL of detection antibody, incubate at 37°C for 1 hour |
| 6 | Wash 3 times, add TMB substrate, incubate at 37°C for 15 minutes |
| 7 | Add stop solution, read absorbance at 450nm |
Sample Preparation:
Collect serum, plasma, or saliva samples using standard protocols. Centrifuge blood at 2000×g for 15 minutes at 4°C. Saliva samples should be centrifuged to remove debris. Store at -20°C. Avoid repeated freeze-thaw cycles.
Storage Conditions:
Store at 2-8°C. Do not freeze. Kit is stable for 6 months when stored properly.
