| 描述 | Tenofovir hydrate (GS 1278 hydrate) is a nucleotide reverse transcriptase inhibitor with antiviral activity, inhibits EBV replication, and is used in the study of HIV and HBV. |
| 体外活性 | Tenofovir hydrate 在 HK-2 细胞中表现出细胞毒性作用，在 MTT 测试中，48 小时和 72 小时时的 IC50 值分别为 9.21 和 2.77 μM。Tenofovir hydrate 降低了 HK-2 细胞的 ATP 水平，并在 3.0 至 28.8 μM 范围内增加了氧化应激和蛋白质羰基化。此外，Tenofovir hydrate 通过线粒体损伤诱导 HK-2 细胞凋亡。[1] Tenofovir hydrate和 M48U1 在 0.25% HEC 中的配方可以抑制 R5 嗜性 HIV-1BaL 和 X4 嗜性 HIV-1IIIb 在活化的 PBMCs 中的复制，同时也能抑制多个实验室菌株和来源于患者的 HIV-1 株。M48U1 和 Tenofovir hydrate 的联合配方在对抗 R5 嗜性 HIV-1BaL 的感染中表现出协同的抗病毒活性，对 PBMCs 无毒性。[2] |
| 体内活性 | Tenofovir Disoproxil fumarate (20, 50, 140, or 300 mg/kg) administered to BLT mice, displays dose dependent activity during vaginal HIV challenge in BLT humanized mice. Tenofovir Disoproxil fumarate (50, 140, 300 mg/kg) significantly decreases HIV transmission in BLT mice [3]. Tenofovir Disoproxil fumarate (0.5, 1.5, or 5.0 mg/kg/day, p.o.) induces a dose-dependent decline in serum viremia in woodchucks chronically infected with WHV. Tenofovir Disoproxil fumarate administration is safe and effective in the woodchuck model of chronic HBV infection [4]. |
