| 描述 | NIK SMI1 is an effective and selective NF-κB inducing kinase (NIK) inhibitor. It also inhibits NIK-catalyzed hydrolysis of ATP to ADP (IC50: 0.23±0.17 nM). |
| 体外活性 | NIK SMI1在HEK293细胞中抑制NIK SMI1反应元件调控的萤火虫荧光素酶报告基因的表达（IC50：34±6 nM）。NIK SMI1还能抑制BAFF诱导的小鼠B细胞体外存活（IC50为373±64 nM），符合NIK抑制剂的预期效果，NIK SMI1被证实能够抑制p52（RelB）的核内转移（IC50=70 nM）。 |
| 体内活性 | NIK SMI1的药理学研究已在SD大鼠、CD-1小鼠、比格犬及猕猴上进行，其给药速率分别为20、32、18及7.8 mL/kg每分钟。分布体积(Vd, L/kg)分别为1.35、1.58、0.778及1.39。在7天的实验期间，C57BL/6小鼠每天两次口服NIK SMI1，或接受三次重组BAFF受体融合蛋白(Br3-mIgG2a)的注射作为阳性对照。 |
