| 体外活性 | FPFT-2216（1 μM；5小时），不仅能在MOLT4细胞中降解其已知靶点IKZF1、IKZF3和CK1α，同时还显示出PDE6D的降解效果[2]。FPFT-2216（1 μM；0小时、2小时、4小时、6小时、16小时、24小时）在2小时内显示出PDE6D的完全降解，并且在MOLT4细胞中PDE6D的降解效果至少持续24小时[1]。FPFT-2216（0 nM、1.6 nM、8 nM、40 nM、200 nM、1 μM；4小时）在8 nM剂量下展示出超过50%的PDE6D降解率，而在200 nM剂量下观察到PDE6D、IKZF1、IKZF3和CK1α的最大降解率[2]。FPFT-2216不会阻碍依赖KRASG12C的MIA PaCa-2细胞的生长[1]。FPFT-2216（10、20、40 μM；14或24小时）显著上调IL-2表达，尽管其在naive CD4+ T细胞中的效果不如Pomalidomide[2]。FPFT-2216（10 μM；14或24小时）能在naive CD4+ T细胞中降解免疫调节药（IMiD）的泛素-蛋白酶体降解底物IKZF1和CK-1α[1]。 |
