| 体外活性 | Zanamivir降低致死测试小鼠模型中病毒的肺滴度并降低发病率和死亡率. |
| 体内活性 | Zanamivir在体外有效降低病毒的复制。Zanamivir抑制流感神经氨酸酶,并防止唾液酸残基的裂解,从而干扰粘膜分泌物中的子代病毒传播并降低病毒感染性。Zanamivir还减少由神经氨酸缺陷型变体形成的噬斑的面积,证明其对细胞-细胞融合的干涉与抑制神经氨酸酶的活性无关。Zanamivir可防止持续感染细胞与未感染细胞的血细胞吸附和融合。通过脂质混合和内容物混合来判断,Zanamivir对血细胞吸附没有影响,但确实抑制HA2b-红细胞融合。Zanamivir导致流感A/H1N1变体His274Asn以及His274 gly,His274Ser和His274 gln的灵敏度降低。 |
