One important factor to consider when selecting antibodies is clonality. Clonality depends on whether the antibody comes from different B cells of parental clones (polyclonal antibodies) or the same B cell (monoclonal antibodies). These antibodies each have their own advantages and limitations. Polyclonal antibodies are a mixture of different antibodies, originating from the immune responses of multiple B cells, and each antibody can recognize different epitopes on the same antigen. Because polyclonal antibodies can detect a range of epitopes on a protein, they carry a higher risk of batch-to-batch variability compared to monoclonal antibodies. However, since polyclonal antibodies are not specific to a single epitope, they are more tolerant of minor changes in the antigen compared to monoclonal antibodies. Monoclonal antibodies are produced by a single B cell parental clone and therefore can only recognize one epitope of an antigen. Monoclonal antibodies are highly specific to the targeted antigen epitope, have low non-specific cross-reactivity, and exhibit very little batch-to-batch variation.

In theory, the species of the sample to be tested should be different from the species of the antibody. For applications such as western blot that use cell lysates without endogenous immunoglobulin (IgG), or experiments that use directly labeled primary antibodies, this issue does not need to be a concern.
