（1）将培养24小时左右的枯草芽孢杆菌或其他芽孢杆菌，作涂片、干燥、固定。
（2）滴加3—5滴孔雀绿染液于已固定的涂片上。
（3）用木夹夹住载玻片在火焰上加热，使染液冒蒸汽但勿沸腾，切忌使染液蒸干，必要时可添加少许染液。加热时间从染液冒蒸汽时开始计算约4—5分钟。这一步也可不加热，改用饱和的孔雀绿水溶液（约7．6%）染10分钟。
（4）倾去染液，待玻片冷却后水洗至孔雀绿不再褪色为止。
（5）用番红水溶液复染1分钟，水洗至水为无色。
（6）待干燥后，置油镜观察，芽孢呈绿色，菌体呈红色
