LMBR1L (Limb Region 1 Protein Homolog-Like) is a transmembrane protein that plays a crucial role in immune system regulation and Wnt signaling modulation. It is essential for lymphocyte development, functioning as a negative regulator of the canonical Wnt signaling pathway. LMBR1L works in concert with UBAC2 and the E3 ubiquitin-protein ligase AMFR to promote ubiquitin-mediated degradation of β-catenin (CTNNB1) and the Wnt receptors FZD6 and LRP6, thereby stabilizing the β-catenin destruction complex necessary for maintaining proper CTNNB1 levels. In addition to its signaling functions, LMBR1L also serves as a receptor for lipocalin-1 (LCN1) and is capable of mediating its endocytosis, linking it to processes involving cellular uptake and trafficking.
