Sequestosome-1 (SQSTM1/p62) is a multifunctional adaptor protein integral to cellular homeostasis. Encoded by the SQSTM1 gene, p62 facilitates the degradation of polyubiquitinated proteins by linking them to autophagosomes and proteasomes, thus playing a pivotal role in selective autophagy and the ubiquitin-proteasome system. Beyond its role in protein turnover, p62 acts as a signaling hub, modulating pathways such as NF-κB and mTOR, which are crucial for inflammation, cell survival, and metabolism . It also contributes to bone remodeling by regulating osteoclastogenesis. Mutations in SQSTM1 have been linked to various diseases, including Paget's disease of bone, neurodegenerative disorders like ALS and frontotemporal dementia, and certain cancers . Due to its central role in critical cellular processes, p62 is a significant focus in research on autophagy, aging, and disease pathogenesis.
