Jagged1 (JAG1) is a transmembrane protein that serves as a ligand in the Notch signaling pathway, a highly conserved mechanism governing cell fate decisions during embryonic development and tissue homeostasis. Encoded by the JAG1 gene located on chromosome 20, Jagged1 interacts with Notch receptors on adjacent cells, initiating a cascade of proteolytic cleavages that release the Notch intracellular domain, ultimately influencing gene transcription. This signaling is pivotal in various developmental processes, including organogenesis and vascular development. Mutations in JAG1 are associated with Alagille syndrome, an autosomal dominant disorder affecting the liver, heart, and other systems. Additionally, aberrant Jagged1 expression has been implicated in tumor progression and poor prognosis in certain cancers.
