G protein-coupled receptor 146 (GPR146) is an orphan receptor encoded by the GPR146 gene in humans. It is predominantly expressed in adipose tissue, with additional expression in the liver, heart, and lungs. GPR146 plays a significant role in regulating plasma cholesterol levels. It serves as a receptor for cholesin, a gut-derived hormone released in response to dietary cholesterol absorption. Upon binding to cholesin, GPR146 signaling inhibits cholesterol synthesis. Studies in mice have demonstrated that genetic disruption of GPR146 leads to lowered serum cholesterol levels and a reduction in atherosclerotic lesions, suggesting its potential as a therapeutic target for hypercholesterolemia and atherosclerosis.Additionally, GPR146 has been implicated in pulmonary vascular remodeling by promoting pulmonary artery smooth muscle cell proliferation through the 5-lipoxygenase pathway. Overall, GPR146 is emerging as a critical regulator of lipid metabolism and vascular function, making it a promising candidate for therapeutic intervention in cardiovascular diseases.
