Twist1 is a basic helix–loop–helix (bHLH) transcription factor encoded by the TWIST1 gene, playing a pivotal role in embryonic development and various physiological processes. It regulates gene expression by forming homo- or heterodimers that bind to specific DNA sequences, influencing the transcription of target genes. During embryogenesis, Twist1 is essential for mesoderm formation, neural crest cell migration, and the development of structures such as bones, muscles, and the craniofacial skeleton. Mutations in TWIST1 are associated with Saethre–Chotzen syndrome, a genetic disorder characterized by craniosynostosis and limb abnormalities. Beyond development, Twist1 contributes to processes like epithelial–mesenchymal transition (EMT), cell differentiation, and immune response modulation. Its involvement in EMT links it to cancer progression, where it promotes tumor cell invasion, metastasis, and resistance to therapy.
