Phospho-eIF2α (Ser51) is important in the control of protein synthesis during cellular stress responses. eIF2α is a core component of the eIF2 complex, which is essential for the initiation of translation in eukaryotic cells. Under stress conditions—such as endoplasmic reticulum (ER) stress, amino acid deprivation, viral infection, or oxidative stress—specific kinases (e.g., PERK, GCN2, PKR, and HRI) phosphorylate eIF2α at Ser51. This phosphorylation inhibits global protein translation by preventing the recycling of eIF2-GDP to eIF2-GTP, thereby conserving energy and reducing the accumulation of misfolded proteins. At the same time, it selectively promotes the translation of specific mRNAs, such as ATF4, involved in the integrated stress response (ISR). Due to its central role in regulating translation and stress adaptation, Phospho-eIF2α (Ser51) is a key biomarker and target in studies of neurodegenerative diseases, cancer, viral infections, and metabolic disorders.
