​Toll-like receptor 4 (TLR4) is a transmembrane protein that plays a pivotal role in the innate immune system by recognizing pathogen-associated molecular patterns (PAMPs), such as lipopolysaccharide (LPS) from Gram-negative bacteria, and damage-associated molecular patterns (DAMPs) released from injured tissues. Upon activation, TLR4 initiates intracellular signaling cascades through adaptor proteins like MyD88 and TRIF, leading to the activation of transcription factors such as NF-κB and AP-1. This results in the production of pro-inflammatory cytokines and type I interferons, facilitating the body's defense against infections and contributing to inflammation. TLR4 is predominantly expressed on immune cells like monocytes, macrophages, and dendritic cells, but is also found on non-immune cells, including epithelial and endothelial cells. Beyond its role in host defense, TLR4 is implicated in various pathological conditions, including sepsis, cancer, and neurodegenerative diseases, due to its ability to mediate inflammatory responses to both microbial and endogenous stimuli.
