​Caveolin-1 (CAV1) is a 21–24 kDa integral membrane protein that serves as the principal component of caveolae—flask-shaped invaginations of the plasma membrane found in many cell types, including endothelial cells, fibroblasts, and adipocytes. It plays a pivotal role in various cellular processes such as endocytosis, transcytosis, cholesterol homeostasis, and signal transduction. Caveolin-1 interacts with numerous signaling molecules, including Src family kinases, endothelial nitric oxide synthase (eNOS), and components of the Ras-MAPK pathway, thereby influencing cell proliferation, apoptosis, and differentiation. Notably, CAV1 exhibits dual roles in cancer biology; it can function as a tumor suppressor in early stages of cancer development, while its re-expression in advanced stages is associated with tumor progression, metastasis, and multidrug resistance. Additionally, caveolin-1 is implicated in the regulation of cellular metabolism and has been linked to various diseases, including pulmonary arterial hypertension and atherosclerosis, underscoring its significance in both physiological and pathological contexts.
